More about PT and PD

As you are most likely aware in many studies regarding Ginseng, Ginseng contains the two main saponin groups:

PTs (panaxatriol) and PDs (panaxadiol), divided according to where and how many of the sugar moieties are linked. The number of sugar moieties and the location, determine the hydrophobicity (or lipophillicity), the three dimensional compliance of ginsenosides with certain receptors. Altogether, these characteristics bring the main pharmacological efficacy and specificity of ginsenosides.



 

Further, the ratio of PD/PT becomes the more important issue in the practical perspective due to the ratio being higher,  it is assumed to be a Suppressant(Yin action) rather than a Stimulant(Yang action). Generally speaking, Korean Ginseng¡¯s PD/PT ratio is only 1.5, but if it is processed to Black Ginseng, it grows to be more than 7. But this ratio also has a limitation to explain fully about true nature of ginsenosides.



 

The trace amounts of rare ginsenosides found only in Red Ginseng and wild mountain Ginseng like Rg3 and Rg5 are becoming more prevalent ingredients resulting in an interesting phenomenon in processed Ginseng products. These constituents also belong to the PDs family of ginsenosides.



 

The First Hypothesis:



 

PT  = A Stimulant           - A Yang Agent

PD  = A Suppressant      - A Yin Agent

Based on my support and research of Ginseng and its properties, I developed the following hypothesis:



 

PT = A Stimulant, A Yang agent. PD = A Suppressant, A Yin agent. There were many clues that supported my hypothesis about the ¡°true¡± nature of Ginseng and ginsenoside. Please allow me to explain:



 

First, although White Ginseng, Red Ginseng and Black Ginseng contain a variety of saponin, we can categorize it into two sub-units and define its action, PT as a Stimulant, and PD as a Suppressant. Actually, the meaning of the Stimulant and Suppressant are focused on the direction not on the benefit it gives. The simulant hereto is an agent that stimulates initial response in metabolism on the level of a cell, organ or system in an up-regulating manner. The Suppressant has the opposite process, meaning in a down-regulating manner. When it comes to the disease process, the Stimulant action on malignant cells is not beneficial to our body physiologically, and in the same manner, the Suppressant action on the malignant cell function could lead in good result to the human body.



 

For example, one of the typical PD, Rg3 acting as an angiogenesis inhibitor, has a strong Suppressant character. Another Rb1 which is abundant in North American Ginseng acts as a general Suppressant at all levels of biological functioning.



 

 In contrast, the representative ingredient of Korean Ginseng Rg1 is a main component of PT type ginsenoside which acts on the glucocorticoid receptors to induce angiogenesis acting as a Stimulant in the human body.



 

As every rule has exception, in particular situation the a certain ginsenoside(the intermediate polarized ginsenoside) has dual mechanism to be both Stimulant and Suppressant . Comprehensively, it is reasonable to assume that PTs are Stimulants and PDs are Suppressants.



 



 

The Mechanism of Selectivity and Efficacy of Ginsenosides



 

The Molecular polarity of the PDs is relatively more hydrophobic (or lipophilic). And with the help of lineage structure , PPD, the aglycon of the PDs tends to permeate the cell membrane to bind inner cell receptors which initiate a m-RNA transcription, a fundamental and structural change. Whereas the PTs family branched at C-6 position goes through a relatively shallow pathway binding surface membrane receptors and causes a faster response and increases the secretion of the short acting hormonal peptide or cytokines that stimulate other cells, a short term and functional change. Anyhow, the selectivity and efficacy of ginsenosides vary in a large range according to its polarity and structure.



 



 



 

When we take Ginseng products, the two opposite co-existing components act their own properties as stimulatory PTs and suppressive PDs respectively. However, stimulatory effect usually comes first ahead of any suppressive effect in a biological process and response. We may experience stimulatory effects of PTs first. This short onset of PTs action can make some people complain about the commonly known side effects of Ginseng. The side effects which often appear after taking Ginseng products (i.e. hot flashes, insomnia, tachycardia, etc.,) may be due to the response initiated by consecutive immuno-stimulatory cytokines in the level of immune responsible cell activity. This also seems to be one of the typical Stimulant effects of PTs dominant ginsenosides.



 

Additionally, the contradictory action of Ginseng is often called Paradox of Ginseng action. It is very common that depending on the constitution of individuals and species of Ginseng, the effectiveness of Ginseng products varies so much that even the experienced practitioner could not predict perfectly. 



 

Among the Ginseng products, White Ginseng and Red Ginseng are assumed as PTs dominant products and the most paradoxical. Although there are PDs family ginsenosides, their potential bioactivity is less powerful until the dominant ratio is above a certain number, it is not clear but one interesting formula gives me an insight to determine the degree of bioactivity of each ginsenoside and the ¡°Yin and Yang¡± of the Ginseng product. Most importantly, the paradoxical effect of a Ginseng can be predictable with this formula.



 

Second Hypothesis:  Bioactivity, Index of Yin/Yang, Index of paradox



 

1)        The bioactivity of the PD family is determined by



 

 BA (Bioactivity) of PD ginsenoside = Detected ginsenoside mass x retention time ratio



 

 The total BA PDs = Sum (ginsenoside mass x retention time ratio)

The maximum retention time = (usually) Rd retention time x 2



 

To reach this conclusion, I considered many factors that can affect the bioactivity of ginsenoside. Besides the mass of ginsenoside, the two main factors (the polarity of ginsenoside and structural affinity) were integrated into retention time. Therefore, to determine the total bioactivity, the retention time and its ratio on the HPLC diagram was adopted.



 

As for PDs ginsenosides, the more distant it is from the start point, the more lipophilic it would be and the more powerful

efficacy would be expected.  I have never failed to find this tendency in my research. Thus, if we have Ginseng that shows a distant ginsenoside profile on the diagram of HPLC, there must be more possibilities to find more powerful ginsenosides.  I found that Black Ginseng has a perfect ginsenosides profile with a high bioactivity, a high potent, a less paradoxical index and PDs dominant ginsenosides.



 

2) The bioactivity of PTs family is determined by ,

 BA (bioactivity) of PTs ginsenoside = detected ginsenoside mass x retention time ratio) 



 

The total BA PTs = Sum (detected ginsenoside mass x retention time ratio)

The bioactivity of PTs family is calculated separately ,because its tendency is opposite of PDs.

The bioactivity of PTs family ginsenosides is also increased with the retention time ratio, they should be summed up as a Yang.

 

3) Index of Yin/Yang Tendency = Sum (BA PT) - Sum (BA PD)



 

As per above, the subtraction of BA PDS from BA PTS would be the index of Yin/Yang. It would be ideal that the value of

White Ginseng is over zero, but because of experimental imperfection its value goes down below zero. One day

we can find a complementary factor (polysaccharides may be the missing fraction of Yang in this index).

With this value we can decide which is more Yang, and which is more Yin. According to this tendency, we can predict whether a Ginseng product acts as a Stimulant or a Suppressant overall.



 

4) Index of potency = sum (BA PT) + sum (BA PD) - this value tells us that although the saponins total mass decreased, the potential efficacy may increase. As the Ginseng is processed, the potency of Ginseng would not decrease because the more powerful ginsenosides will be increased.



 

5) Index of paradox

= {sum (BA PT) + sum (BA PD)} / ABS {sum ( BA PT ) - sum (BA PD)}



 

Paradoxical effects are increased when this value grows. With this index, the Ginseng products are evaluated for its clinical adaptability and can be predicted the possibility of side effects after consuming it.

From this standpoint, Black Ginseng is the least paradoxical Ginseng. White Ginseng is the most paradoxical Ginseng. It means the effect of White Ginseng is not predictable although its potent is high. Conversely, as this value gets smaller, the specificity of the Ginseng product will increase. Clinically, the least paradoxical Ginseng can be the best candidate for pharmacological use.



 

In conclusion, with this formula it is easy to understand how a Ginseng product will act, whether it will be a Stimulant or a Suppressant. We also know the total potential efficacy and the quality of a Ginseng by measuring the index of potent and the index of paradox respectively. By comparison with each Ginseng product before applying and administering it to a patient, we can avoid the potential side effect of the Ginseng product. In other words, the Yin deficient patient would be better to take a Yin oriented Ginseng, and the Yang deficient patient, a Yang oriented Ginseng. In the future we could find a way to fortify the tendency of Ginseng or neutralize it to customize for the individual¡¯s needs and health condition.



 

Please note that I have attached an Excel file that outlines my assumption perfectly as it coincides with the experiment of two cases that analyzed the changes of ginsenosides according to the steaming times in Ginseng. I think this formula and hypothesis will act as a very powerful tool in evaluating Ginseng products and providing us with the intuition regarding Ginseng and how to use them properly.
